House has locked onto the brilliant combination of what I like to call "series plot" and "cartoon plot".
A series plot is an on going plot that over time defines the characters, and follows their lives. This is important because it gives the audience a chance to fall in love with the character's personalities and minds. Also, every new episode that you watch is enhanced by the knowledge that you received in prior episodes making you feel more connected to the characters. Other "series plot" shows include Desperate Housewives or 24.
What makes House great is that it has also brought in "cartoon plot". "Cartoon plot" is when a show has the same characters in every episode but each episode is basically unconnected to another. And example of this would be the Simpson's. While some parts of an episode rolls over to another, like a death or marriage, most of the plots are unconnected. In one show, the family car might blow up but in the next episode it could be good as new.
House brings in this plot with it's patients. Every episode you have new patients to work with. New mystery diseases. This makes the show exciting and addictive. Even if someone hasn't been watching House regularly, they can still enjoy the show purely for the medical drama of diagnosing the patient.
With this perfect blend of "series plot" and "cartoon plot" House has easily won the hearts of many fans. The diversity in personality of the characters is refreshing and each one is well developed and interesting.
House is in my opinion, one of the best shows on TV at the moment.
